if a fire spells deals this talent`s effect on a creature (with 25 spell power) and that creature`s defense get decreased with disruption ray on expert darkness magic after the effect will it be like:
defense is 50 after the effect it get`s 25 and then uses ray it will be 20?
or it will be 50 minus 5 from the ray minus 50 percent=22.5~23?
so,does the dominion effect is on the creatures defense at the moment the fire spell is used or it effects it`s all-the time defense (before it runs out)

I have never used the dominion of fire talent, but judging from the description I believe the 2% per spell power affects only the current defense of the stack and not the original defense.
So then the order in which you use fire and disruption would matter.

The talent description says 1 combat turn. | Looks like it is based on the Defense of the unit at the time that the Fireball hits.

Watch the Hydras and Minotaur Soldiers on the middle of the right hand side of the screen. Before the Fireball they had both defended going from defense 33 to 43 for the Hydra, and from 23 to 30 for the Minotaur Soldiers. After the Fireball hits (with Dominion of Fire) the defense becomes 21 for the Hydra and 15 for the Minotaurs (Wizard has 25 SP for maximum 50% reduction).
Since the Defend option is only a temporary effect it *should* act in a similar fashion to Disruption Ray, which is also, essentially, a "temporary" effect, since it isn;t in place at the start of the battle.
You will notice that when the Dominion of Fire effect expires, the Hydras and Minotaurs return to their previous "Defend" values.
